BIOENERGETICA MITOCONDRIAL
Nuestro interés es sobre el estudio de la bioenergética mitocondrial en dos modelos: la placenta de humano, y el basidiomiceto Ustilago maydis. En los últimos años, nos hemos enfocado en el estudio de los supercomplejos de las mitocondrias de Ustilago maydis, particularmente los dímeros de la F1F0-ATP sintasa y los supercomplejos conformados por los complejos respiratorios I, III2 y IV. Se ha demostrado que los dímeros del complejo V participan en el plegamiento de las crastas mitocondriales. Respecto a los supercomplejos, su composición puede ser III2:IV, I:III2, o I:III2:IV siendo éstos últimos reconocidos como respirosomas, gracias a su capacidad de oxidar al NADH y reducir al oxígeno.
Hemos aislado estos supercomplejos y los hemos caracterizado cinéticamente. Ahora contamos con mutantes de las subunidades dimerizantes del complejo V y hemos iniciado el estudio de la repercusión de la eliminación de las subunidades dimerizantes en la estabilidad del dímero. Adicionalmente, estudiamos la arquitectura mitocondrial en las mutantes de las subunidades dimerizantes.
